Description

Melatonin is synthesised in the pinealocytes, or cells of the pineal gland, from serotonin, through several steps controlled by various enzymes. Melatonin synthesis and release follow a marked circadian rhythm, with peak secretion occurring at night. This circadian rhythm in melatonin production gives the body valuable information about the time of day and the time of year. In mammals, the rate-limiting enzyme in melatonin synthesis (N-acetyltransferase) is under the control of noradrenaline. Melatonin has a clear sleep-inducing action and is naturally a hormone whose levels rise at night and fall during the day. Given its high lipophilicity (it crosses the blood-brain barrier easily) and its non-toxic nature, melatonin may be an important molecule in the brain. In humans there are wide variations in melatonin production over the course of life (in addition to the daily variations already noted): levels are highest in childhood, then decline at puberty and finally fall considerably in adult life.
